Improved production of human hemoglobin in yeast by engineering hemoglobin degradation

چکیده

With the increasing demand for blood transfusions, production of human hemoglobin (Hb) from sustainable sources is increasingly studied. Microbial an attractive option, as it may provide a cheap, safe, and reliable source this protein. To increase by yeast Saccharomyces cerevisiae, degradation Hb was reduced through several approaches. The deletion genes HMX1 (encoding heme oxygenase), VPS10 receptor vacuolar proteases), PEP4 proteinase A), ROX1 heme-dependent repressor hypoxic genes) overexpression HEM3 porphobilinogen deaminase) AHSP alpha-hemoglobin-stabilizing protein) — these changes improved production. validated bilirubin biosensor. During glucose fermentation, engineered strains produced 18% intracellular relative to total protein, which highest reported in yeast. This increased accompanied with oxygen consumption rate glycerol yield, (we speculate) yeast's response rebalance its NADH levels under conditions limitation protein-production.

Degradation of human hemoglobin by organic C-nitroso compounds.

The crystal structure of the nitrosomethane adduct of human Hb shows N-binding of the MeNO ligands to heme Fe. The structure of the EtNO adduct reveals a surprising 4.9 Å heme slippage in the β subunit, and explains the ability of C-nitroso compounds to degrade Hb removing it from circulation.

Degradation of native human hemoglobin following hemolysis by Prevotella loescheii.

Prevotella loescheii PK1295 can grow on native hemoglobin as a source of heme. Supernatants of P. loescheii cultures hemolysed human erythrocytes and degraded native hemoglobin. These combined activities may provide heme (or iron) for the growth of P. loescheii and other dental plaque bacteria.

Inhibition of hemoglobin production by transferrin-gallium.

Recent clinical trials evaluating gallium nitrate as a chemotherapeutic agent have reported the development of microcytic hypochromic anemia in patients treated with this agent. Because gallium is known to bind avidly to transferrin, we examined the effect of transferrin-gallium (Tf-Ga) on hemoglobin production by Friend erythroleukemia cells in vitro. Cellular hemoglobin production, as assesse...

Plasma substitutes; human and animal globin related to the production of hemoglobin and plasma protein; dog hemoglobin utilization improved by methionine but not by isoleucine.

Globin makes up 95 per cent of the hemoglobin molecule. Its construction is of fundamental importance. It is a protein and its production in the body relates to the building stones of protein and to other body proteins. In fact plasma protein introduced into a fasting anemic dog will bring about a large output of new hemoglobin (globin). Globin production and the related hemoglobin can be contr...

Pregnancies Complicated by Hemoglobin H disease

Dear Editor-in-Chief The recent report on “The Adverse Effects of Pregnancies Complicated by Hemoglobin H (HBH) Disease” is very interesting (1). Rabiee et al. reported a pregnant case complicated with HBH disease. Indeed, this problem might not common in the Middle East but it is very common in Southeast Asia. The authors hereby would like to share the experience on this top...
